I am working on a quantitative method using the LC-MS 1100. My analyte is 3-methyl,2-aminopyridine, and it has very poor ionization in the mass spec. Does anyone know of a column, buffer or solvent system that will improve my ionization of this compound? I'm currently using a gradient, 10-95% Acetonitrile + 0.1% formic acid and my DAD detection is good, but my analyte shows up in the baseline of the MS and s/n is very poor. Any suggestions would be greatly appreciated.

Can you inform me about MS conditions: probe voltage, temperature and elluent flow rate. Are you
using SIM mode? Judging from your pH you are producing dication m/2. Perhaps it will be better to use higher pH so that ionize mono amino group and detect monocation m/1. In the higher molecular mass region the noise will be lower.
