Dear all:

A GC-MS method for assaying a drug in plasma (0.2-10 ng/ml)uses liq-liq extraction (heptane) from alcalinized plasma and back extraction in methanol/HCl 0.05N. The acidic extract is concentrated to dryness and the residue is resuspended in 40 ul of dimethylsolfoxide (DMSO). The injection is splitless 1.5 ul, on a 15m x .25 mm x 0.1 um DB5ms, Temp. prog. 170ºC to 220 (40ºC/min) then to 260 (15ºC/min), detection SIM at m/z 342
The problem is that 10 ul of such DMSO extract is used for metabolite assay by HPLC-FLD, therefore we need a water miscible solvent.
Do you have any experience using this solvent?
I've got some doubts about condensation due to its high boiling point.
Any comment would be really appreciate.
Regards

Marcelo
