I am about to start global protein profiling by MS (proteomic analysis) after clean-up/enrichment and groping by SPE, i.e. non-charged proteins by reversed-phase SPE, positively-charged proteins by cation-exchange SPE and negatively-charged proteins by anion-exchange SPE.
Any previous experiences, please advise.
I don't need to separate all proteins like in most of the LC-MS works. Instead I need good SPE cartridges to retain certain groups of proteins for later elution followed by MS analysis. I believe the important parameters will be: sample pH, SPE functional group and pore/particle size, rinsing buffer and eluents. The selection of SPE cartridges is the key.

I'm not personally experienced in this but as they say "I know a man who is" and I've asked him the question. From what you've described as a method development strategy you could try a multi-mode cartridge from IST. It contains C18, strong cation exchanger and a strong anion exchanger all in the one cartridge. I've seen somthing about the separation of acid basic and neutrals on a cation and hydrophobic column as well.
I agree that pH and pore size will be critial. Have you thought of using a array to do method development.
