I want to know exact mathematical way for calculating matrix effect of a plasma for any particular drug analysis, using lc/ms/ms.

One way is described in J. Mass Spectrom., 2003, 38, 823-838.

In summary: spike the same amount of analyte into solvent and blank plasma extracts, and ratio the peak areas.

There was also an article in Anal Chem June last year. The technique is simple, as described by MG. Preferentially, you do not want to have any matrix effects, since the matrix may be variable from sample to sample.
