Developping an LC method, I observed that only DMSO as organic modifier lead to a sufficiently good method. I wonder if this solvent can well be used if I furhter want to couple LC to MS?

I don't know how DMSO behaves as far as getting good ionization. I've never used it in my mobile phase. But there's another issue that could present a problem. Some ion source designs use PEEK tubing as the transfer line between the ESI needle (at ~ 5kV) and a grounded union, for safety. I think PEEK is incompatible with DMSO. Replacing that particular length with stainless steel would short the power supply.
