I have very little experience with SEC/GPC assays, and I need to develop a method that will verify MW of low-MW dextran. The target sample MW is 1000, and a test is desired to verify the mean MW is within 900 - 1100. I have a method that uses several in-series columns and a run time of several hours. Is a single-column SEC/GPC LC assay a realistic goal? I have seen literature demonstrating the separation of dextrans over a large MW range using a single column, but none that are as specific as I would like. Any suggestions?

Select a column with the target MW in the middle of the calibration curve. Then buy as many columns of the same type that you feel you need to get sufficient resolution. If you buy too many, you got spares for the future.
Based on your description, one should be enough...
