However I changed mobile phase from 5%--80%(Meth:Water)I receive only a peak when injecting sample.
Could I increase ph?How to adjust?Could adjustment hurt ODS colume?

You can vary the pH from 2 to 8, no problem, and you probably should control the pH. I don't know what your compounds are. Can your detector see them?

I am very interested in your application for matrine and related alkaloids.

First of all, I also have doubt (as rised by Uwe Neue) on your detection mode. Since matrine doesn't come with significant UV chromophore, we prefer to use GC-MS for sensitive applications.

Probably if you are usually UV detection, it worths doing a preliminary experiment to scan with a UV spectrophotometers your standard solutions (individually diluted in your HPLC mobile phase), to ensure sufficient detectability and select optimal wavelengths, etc.

By the way, I would like to know where did you get these reference compounds from? since I am searching for reliable suppliers of those herbal compounds but not very successful.

I've tried GC-MS and detected it.but,as U know,if sample goes through MS,it's hard to detect by quantity.So I give up this way.I try to vary mobile pharse PH to detect and it's not excellent.
