We are currently attempting to qualify the current BP/EP HPLC method for the assay of Erythromycin base.
In short we have found the method to be practically unworkable from even such basic chromatographic requirements such as mobile phase, pH stability, column temperature and even to tubing ID !
Can anyone help with advice on HPLC alternatives or hints if you have worked with the same material.

Hello, I am sorry to respond to this message two months after you posted it (I just got internet access). I have a paper which lists about 40 references for HPLC methods of erythromycin analysis. I haven't read it, but most of them appear to be pretty simple - C18 columns and buffer/organic mobile phases. The paper is Journal of Chromatography A, 812 (1998) 255-286. I hope this is helpful.

Phenomenex supplied me with a rather easy application on a Prodigy C18 columns. It is a gradient method. I dont remember the exact conditions off hand but they should be able to supply you with the method.
