I found a literature reference to a separation that I would like to perform. The mobile phase used in the reference is a 25 mM phosphate buffer/acetonitrile gradient. Could anyone give me a procedure for preparing the 25 mM phosphate buffer? Also from the way it was stated in the reference, I assume the buffer is only added to the water and not to the acetonitrile. Is this correct? Thanks in advance for your assistance.

Dissolve 25 mmole (FW x 0.025) phosphate in 1 L water. Adjust pH if necessary. If the method has pure MeCN as the B mobile phase, there definitely is no phosphate in it as it won't dissolve.
