I am looking for a procedure that will separate a mixture of phosphotidylcholines based on their fatty acid chain lengths, e.g. separation of DMPC, DPPC, DSPC, etc. A reverse phase techniques seems natural for analytes differing in hydrocarbon chain length; however, I'm not sure reverse phase is appropriate due to the zwitterionic nature of the polar head group. Any suggestions would be greatly appreciated.

you can do a RP separation for these phospholipids. I have found good separation using an xterra RP8 column, 0.05% TFA in water (A) and methanol (B). Start with around 80% B and go to 100% over 20 minutes. My samples are made up in 98/2 methanol/water. Hope this helps
