Ion pair chromatography

Dear all,

I have gone thru quite a lot of the literature and found that most large antibiotics e.g. gentamicin, erthyromycin, neomycin, etc normally employ ion-pair chromatographic technique for their HPLC separation. Is it a must? Any other option as ion-pair chromatography is not that easy to optimize. Thanks.

There is really not a specific need. It is quite possible that most of those methods are rather old, older than maybe 5 years. Back then, one did not have the range of packings capable of separating compounds at neutral pH as they are avaiable today. On the other hand, why do you think that ion-pairing is more difficult to optimize?
