I'm interested in extracting liposomes from a biological sample and analyzing them for their phospholipid content. I have an HPLC/ELSD method for the analysis of the phospholipid but all the sample prep is in methanol. I need a way to break down the liposome and keep it soluble, the only idea I have come up with is using a roto vap for the samples then reconstituting in methanol and spinning or filtering out the proteins that precipitate out. I don't have a roto vap yet and would like to know if anyone has any ideas or experience with this?

Most likely, the best method for lipid extraction is the method by Bligh and Dyer. See the GC Message Board, "Fatty Acid Analysis + SPE, ~Feb. 20, 2001.
