Hi there!

I want to get some input regarding the choice of wavelenght selection for purity analyses of libraries with LC. The best compromise is probably using a DAD and sum the range of ex. 210-450 nm. If a DAD is not present in the system I suggest to use 220 nm. Some chemists claim that 254 nm is better due to a "cleaner trace" (salt ion etc does not show up).

Any comments? Please!

A single wavelength can not give you an estimate of purity of a peak. As well, a DAD can only really give the purity estimate of an individual peak (ie, are there co-eluting components within that peak).
