A colleague and I had a difference of opinion on mobile phase preparation. The mobile phase in the method is stated as MEOH/H2O (50:50) containing 0,05M B5. If you are using your system in “dial a mix” mode should you make up 0,1M B5 as mobile phase in line B and 100% MEOH in line A. If you leave the concentration of the mobile phase in line B as 0,05M B5 the final concentration of the mobile phase going through the column will be MEOH/H2O (50:50) containing 0,025 M B5. Am I correct in adjusting for the B5 when “dial a mix” mode is used?

I would make up each phase with 0.05% B5, and let HPLC unit mix at 50/50. Let me state that I am a big fan of authors better-detailing stuff like this to eliminate any ambiguities.

I don't see the ambiguity MeOH/H2O 50:50 containing 0.05 M B5, meaning the mixture contains 0.05 mol B5 per 1 L of volume of 50:50 mix. Now YOU need to figure out, how to do this with dial-a-mix mode so that there is not a problem.

It's OK to add 0.1M B5 in water (line B) and methanol without B5 in line A.
