Hello
I have developed and Validated a method in RP using Cyano column. The mobile phase is 85% Acetate buffer:5% THF:5%MeOH:5%CH3CN with less than 1% Butylamine modifier. I am encountering problem with one peak which looses it's resolution very slowly. When I started validating the method, the resolution was close to 3.0 and when I completed my validation it came down to 2.3 or so. I was wondering if this is due to the amine modifier than I use in the mobile phase with cyano column. Can we use amine modfiers when using cyano columns in RP? I know that the tailing reduces with amine modifiers since they help to form ion pairs.

Cyano columns are not very stable. This may be the problem.
