Hi everyone! Does anyone know a good method for calibrating HPLC (autosampler, pump, detector) that takes into account linearity and precision? Any help would be greatly appreciated!

We use the Agilent 1100 OQ/PV as the basis for the calibration of all of our HPLCs. We suplement the procedure with injector linearity, dwell volume, and bandspread tests. Send me an e-mail if you want a copy of our procedure. Good luck.

I have recently contacted Waters Corporation, they have new software (AQT) for calibration of their Alliance HPLC systems, it's based on the retention times of coffeine and Methyl-para-hydroxybenzoaat en their Symmetry columns, for example: when the flow changes from 0.5 to 1 to 2 ml/min, the retention time changes linear, the point on the Y-axis is the fault onto the flow.
Ask Waters about the theory behind their AQT and you will know how it works. Good luck

BJ, Thank you very much for the information regarding the new AQT software. We have many Alliance HPLC Systems at our company. It looks very interesting and hopefully we can apply it in our lab.
