I am having difficulty developing a method for the analysis of DMSO (circa 3%) in water. I wonder if anyone may have some advice on how to proceed. Evidence gained so far, has established that DMSO is the only contaminant.

I have never had need to determine DMSO, but DMSO/water has been a solvent in things I have analyzed. From this experience I know that it can be retained on a Aquasep column (ES Industries) and probably every other "aqueous" column and it gives a good UV absorbance signal. I don't know what the peak shape will be because in my cases it was always overloaded, but I don't see any reason that there would be a problem.

I have been able to assay DMSO using a Zorbax column RX-C8 5 micron. With a mobile phase of 0.01% NaCl, 0.1% H3PO4. It is only retained for 3.5 minutes at a flow rate of 1 mL/min. LOQ is about 0.05%
