The best thing to do is to never let the pressure go up. Use a guard column!

Instead of passing pure ACN or MeOH, better use ACN/water or MeOH/water. If it doesn't help, change the upper frit.

Hi,
what happened to your TSK Super ODS column? In general backpressure for this 2µm column is lower, compared to other 2,5µm materials, even with flow rates of 2ml and ambient temperature.
In our TSK-GEL HPLC Column Instruction Manual we recommend to rinse the column with Acetonitril or Methanol, and run a gradient from 10% to 100% acetonitril in 0,05% trifluoroacetic acid. The Tosoh Biosep OCS sheet can also give you some more information on the TSK Super ODS material. Just e-mail to g.kratz@tosohbiosep.de and I will take care that you will get all information.
On the other side you should give us more information about your chromatographic conditions and your sample. What sample matrix, what sample preparation, what mobile phase preparation etc., all can influence the column performance.
Please give me more information, so we can work on this problem.

The reason that the TSK "2 micron" column has a lower backpressure than other 2.5 micron columns is simply that the particle size is larger, meaning over 3 micron.

I HAVE TO TELL YOU, THAT I´VE BEEN WORKING USING A MICROFILTER, ALWAYS WITH WATER AND MeOH, OR AcN;
THE SYSTEM TO CLEAN THE COLUMN WAS THE NEXT:
-H2O/MeOH IN GROWING % OF ORGANIC DSV.
-H2O/AcN/THF INCREASING % OG ORGANIC.
-H2O/AcN (IDEM)
I´VE BEEN PASSING THIS CICLE SEVERAL TIMES AND BEFORE CHRISTMAS TIME, I HAVE GET ONLY 10 BAR MORE THAN GOOD PREASURE , BUT AFTER THE HOLYDAY , IT WAS MORE THAN 100 BAR UP .
THE CHROMATOGRAPHIC CONDITIONS WERE: AcN/BUFFER OF KH2PO4, AND THE SUBSTANCES WERE BASIC BETALACTAMIC ANTIBACTERIALS.

On our certificate of analysis we publish the particle size of TSK-Gel Super ODS 2µm! It is 2,29µm! And SD is 0,27µm! Thank you very much that you confirmed that the Super ODS 2µm has lower backpressure compared to other 2,5µm materials. And to complete the story: Pore volume is 0,25ml/g, specific surface area is 96,8 m2/g, mean pore diameter is 11,2nm, exclusion limit is 20.000Da, and theoretical plate number is about 200.000N/m!
