Hello,
Sometime ago I read a message about to do a make up after the column like in GC,anyone has more details ?I mean,in GC we use the make up to create a more consistent flame(FID) and have more ions(hitting) in the eletrode .
but in HPLC with UV detection The matter is the UV absorption of my compound ,if I dilute it with a post column make up, it must absorb the same no?

Thanks

Fabiano

Absorbance is proportional to concentration. Therefore if you dilute your sample with post colunm make up, its concentration is less, and its absorbance is less. However, it is possible to do a post column derivitization to increase the absorbance of your compound.
