I am using LC/MS method to determine the concentration of fat acid in urine. I plan to use SPE method to treat get rid of protiens ( not for concentration), but someone suggested that using a biotrap column as a precolumn is better. I have no idea of such column and method, any advise and suggestion will be appreciated, especially about the detail of the column. The HPLC column i am using is C18 1X100nm.

The BioTrap 500 is a very short (2cm) extraction column capable of handling serum or plasma with ease. It is made over in Europe but is available through many companies here in the USA. Search BioTrap on the WWW or simply visit their web-site at www.chromtech.se/biotrap. Two companies which supply the BioTrap in the USA are Analytical Sales and Services (New Jersey) www.analytical-sales.com and Chrom Tech (Minnesota)www.chromtech.com. Chrom Tech, Inc. is not the same company as ChromTech SE.

Just checked the Chrom Tech page, the BioTrap column is just another restricted access column of which quite a few variants exist presently. (The Pinkerton column is on the market well over 10 years).
If you are successful in using this type of column without any other prior clean-up please let us know!
I don´t want to give a wrong impression, thus it is appropriate to say that I am highly appreciating restricted access columns (especially the Pinkerton, still), but manufacturer´s claims?????
Incidentally, when all else fails there is still the good old extraction (see the GC forum on FA, etc.) or contact me.

We have worked out a range of methods for compounds in plasma and urine using LC/MS separations. I recommend to look at C. R. Mallet, Z. Lu, J. R. Mazzeo, U. D. Neue, Rapid Communications in Mass Spectrometry 2002, 16, 805-813. The methods are rather generic and can be applied to a multitude of samples.
