Chiral Separation

Hi everyone:

We have to develop a chiral method for a drug and a metabolite. Both have the same chiral centre but quite different polarity. I have found only literature detailing separation of the drug. My question is: will the column also show chiral recognition for the metabolite?

This is not a chiral separation!!! If the
compounds have differant polarities, and they
are differant compounds, regular HPLC
should separate them!!! Chiral HPLC is for
separating enantiomers, which have the
same physical properties whether R or S. It
does not matter if the drug and it's metabolite
have the same stereochemistry (R and R or S
and S), they are differant chemicals!!!

Do you need to separate the two enantiomers
of the drug? Or the two enantiomers of the
metabolite? This WOULD be a chiral
separation.

We have to separate both the drug and the metabolite on CSP by LC-MS/MS.

What type of CSP do you want to use? My experience on a OD and a AS CSP is that small changes in the physico-chemical properties of the analytes far away from the asymmetric centre require a completely new method. Just try and see!
