Dear Chromatographic users,
Would anyone have ideas/information regarding use of peak purity analysis on peptide molecules analysed using HPLC with a PDA detector. Thank-you for your help.

Check some book, etc., on UV-spectroscopy of proteins (Example the classic: Schultze, Heremans, Molecular Biology of Human Proteins, vol. 1, Elsevier, Amsterdam, 1966). Unless you have very short peptides with widely differing aminoacid constitution (the 280nm chromophore amino acids: tryptophan, tyrosine, phenylalanine) it may be very difficult or impossible.
You may have to use widely differing chromatographic techniques (comparison of RP with IC, etc.) to check into the molecular homogeneity of a peak. MS could help.
