When I inject increasing concentrations of the
peptide I am trying to analyse, the peak starts
eluting a little earlier. The rear (last eluting
part) of the peaks are always aligned.

Can anyone explain this observation?

Thanks.

Brad, you are describing classical overload behaviour. See Snyder and Kirkland's book "Practical HPLC method development"

David

I agree, you may overloading the column. The peak shape will suffer as well.

What are your mobile phase conditions and column? Also, what type of peptide are you injecting and what is the injection solvent?
