Hi,
I have a 1mm C18 15 mm column for the peptides analysis. Once I made a mistake: I use the flow rate of 0.8 mL/min instead of 0.08 mL/min to flush the column, after 10 min, I found its pressure went to 4800 psi, then I stop the pump. Afterwards I found the effeiciecy is terrible. How should I do with it? Does it mean that the column died forever?

At what pressure does your pump shut off?

The max pressure for my pump is 6000 psi.
When I stopped the pump, I disconnect the column so the pressure droped from 3000 psi to 0 suddenly. Will this hurt?

This should not be a problem for a standard well packed column.

Try to backflush your 1mmID column with a flow of 0,01ml/min! Start with 100% acetonitrile, and go 5% stepwise with water, always flushing with 10 column volumns. Than use your column in the reversed direction than indicated on the column lable. It will take some time. Going back to your initial mobile phase and test your column. If column test shows good performance, your column should be ready to use. If not, sorry, but than you must say good by to your column. Also, try to check backpressure while disconnecting your column from your system, to see if your setup gives significant backpressure with 0,8ml/min, or perhaps a capillary connection is blocked. Don't worry, such mistakes can happen. I also learned from such mistakes I did, and today I'm earning my money with selling columns (just kidding). Good luck! Gerhard
