does someone know about an HPLC analytical method [using UV-VIS detector]of DONEPEZIL(ARICEPT)?

Hello Anonymous,
Pfizer uses Inertsil ODS-2 5 µm 150 x 4.6 at 35°C. The mobile phase is prepared as follows: 2.5 g sodium decanesulfonate in 650 ml water. Add 1 ml of 70% perchloric acid and 350 ml acetonitrile. Adjust apparent pH to 2.5. Flow 1.4 ml/min. 271 nm. Inject 20 µl of 0.2 mg/ml donezepil HCl in dilution solvent (0.1 N HCl - Methanol 1:3). Donezepil elutes after appr. 12 min.

Adjust pH after adding acetonitrile? I thought, that we should measure pH in aqueous phase before mixing with organic solvent.
Could you explain this, for I’m new in HPLC, and getting confused.

I admit adjusting pH before adding organic sovent is more staightforward. Anyway, I believe this particular method was developed by adjusting pH AFTER adding acetonitrile (that's why I wrote apparent pH). John Dolan had a very interesting column on this in the November or December 2002 issue of LC GC (a monthly published FREE journal, and available online)!
