I'm told to check the pH of a compound and then pKa for HPLC separation. I have prepared the saturated solution and measured the pH. Is this methodology is correct? Do the compound required to get completely dissolved in water.?

Why titration? pH of filtered soltuion is 5.0
My doubt is , Is it required to completely dissolve by any means, ie can I add some other solvent say methanol , in order to dissolve and then test for pH.

No, don't add solvent. You got the pH.
