Is it a good idea to wash a column with 100% water after running samples with a mobile phase of acetonitrile:0.02%TFA. The gradient starts with 10% acetonitrile to 45%.

Andrea, it is my understanding that washing a column with 100% water will collapse the column packing. You should keep some organic in there, 5-10% min. Then wash w/ a strong organic to restore or stand the packing back-up.

After completion of analysis it is not wrong to
wash the column with the 100% aqueous phase.
then restore the column in organic solvent.
