I'm attempting to analyze the polysorbate 80 concentration in a pharmaceutical matrix (at 50 ppm polysorbate 80). I have decent chromatography, including sufficient sensitivity. The difficulty lies in recovering the polysorbate from the matrix. The matrix contains a relatively high concentration of suspended microcrystalline cellulose (MCC), sodium CMC and dextrose. I have tried: 1) centrifugation to remove the MCC, and the supernatant does not test positive for polysorbate 80; 2) a couple of liquid:liquid extractions without pelleting the MCC, to no avail.
Any ideas would be greatly appreciated. Thank you for your help.
