Hi
I'm a Masters stu. I'm working with purification
of some extra cellular biological substance
produced by yeast. The cell free cultured
supernatant binds strongly ith DEAE-cellulofine
but i failed to elute the substance while using
NaCl (0~2 M) in a test tube method. The activity
of the substance is pH stable from 3~9. I have
used different buffer pH 5.5~8.8 (20mM). What
should i try next to desorb the substance?

Sorry for answering late, thought that someone familiar with cellulofine? would do this. Generally one uses chaotropic substances (see other chains on this), or detergents, or organic solvents (the latter also in concert with either a chaotropic or a detergent)to get proteins in solution.
