Hallo
I will soon go to the wonderful world of biological samples (blood, serum, plasma etc.). I was wondering if you could give some refs or web pages which deal with sample prep. for HPLC. There are tons of papers with informations on this subject, and i would like to fish out the best ones with as much info as possible.

Check at www.waters.com and look for methods using Oasis sample preparation. On-line or off-line plasma, with LC/MS or LC-UV.

I will be developing a method for drug in suction blister fluid, and i am not sure what shall I use as a matrix for blank and spiked samples

Take a look at the Cohesive Technologies. Turboflow columns. These allow for the direct injection of biological material onto the HPLC system without the need for much sample preparation (just a bit of centrifugation and pH adjustment).
For suction blisters you could use serum.

in-line sample preparation can be completed with a dialysis unit. This unit can be used for blood / urine / emulsions / suspensions.
tremendous reduction in time and effort for sample prep ....
the sample is prepared from the sample changer to the injection valve....
