I'm a placement student doing my final year project and it is to try and integrate the separation of Vit B1, B2, and B6 using as few as possible HPLC and mobile phases. Any suggestions?

In a previous work we separated water soluble vitamines like thiamine, riboflavin and pyridoxine on a non-porous column. Run time was 1.3 min, eluent consumption ca. 1.6 cm3:

Try 0.05M KH2PO4 pH 6.5 and a base deactivated column, run a gradient( be aware for precipitation, eg: 5% - 70% )and choose an isocratic % that you think that's suitable and try this. I guess this must give you in a very short time a suitable method.

It appears that your teacher is of the really old school of chromatography. There is an application from 1974 on microBondapak C18 with ion-pair reagents on this separation. If you do not use an ion-pair reagent, you may have a hard time to get them into the same time window.
