I am looking for a HPLC separation of Vitamine B1 TPP and Vitamin B6 PLP in one HPLC run.
Anyone got any suggestions.

Most column vendors have an application library. "water soluble vitamins" is a standard application that is included in most cases.

Stanndard system C18 phase, eluent water+Et3N+heptsulf+acid to ph3 -- acetonitrile.

Try Waters phenyl 300*4.6mm column
with 50 mM hexane 1 sulphonic acid in 1% acetic acid: methanol: acetonitrile(80:15:5) you will get good separation B6 followed by B1.
or use any good c 18.

I've used the Alltech Prevail C18, which is water-wetable. Using 97% phosphate buffer at pH 3.0 and 3% acetonitrile and it has worked quite well. Alltech has an application number 9388 which should help. Try www.alltechweb.com
