Hi all,
I've just taken over the setting up of the High pH (pH9)Agilent Prep LC/MS system at the place I work and I was wondering what conditions, gradients and columns everybody else out there is using. I'm currently setting the system for Low pH to make sure it works properly and I've already had loads of problems,
gradient mixing of ACN/H2O(any cures)
pressure spiking on injection
Pressure spiking into MS using active splitter.]
All thoughts comments and questions welcome.

High pH: XTerra columns, pH 9 to 10 with ammonium bicarbonate (volatile) or ammonia.

For working with high pH mobile phase another choice is Zorbax Extend columns that are stable until pH 11.

Zirconia-based columns also work well at high pH with ammonium bicarbonate/ammonia buffers.
