what is the necessity to dissolve the sample in Mobile phase (as in different pharmacopie)?

A solvent with stronger eluting strength than your mobile phase will act like a strong mobile phase briefly when it hits the head of your column, causing the problems mentioned by anon # 2. Your sample doesn't have to be dissolved in your starting mobile phase, but if not, it should be dissolved in a *weaker* solvent than your starting mobile phase.
