I have 2 methods for you to start with. If these 2 dont work for you I have one more thats a little more complex. I have included one gradient method and one Isocratic. I would recommend the isocratic for only simple matrices
Method 1
Column: Hypersil SAX 15.0 x 4.6 x 5 um
Gradient:
Pump A: 0.005 M Phosphate Buffer pH=2.8
Pump B: 0.750 M Phosphate Buffer pH=3.7
linear curve 100% pump A to 100 % pump B over 20 minutes
Flow: 2.0 ml/min
Detection: UV @ 254 nm
Method 2 (isocratic)
Column :
C18 15.0 x 4.6 x 5 um (Aquasil C18 or equivelent)
Eluent: 0.1 M KH2PO4 pH=6.0 with KOH
flow: 1.0 ml/min
Detection: UV @ 260 nm
