Please can anyone give details about the saponins analysis by HPLC and UV detection. There are several limiting factors for saponins like the chromophore problems, use of low wavelength for detection and hence the drift in baseline etc. I am unable to identify and proceed with quantification of saponins in beans. Please guide.

We have made some preliminary experiments using HILIC mode, and the ZIC™-HILIC columns seems promising for these amphiphilic compounds.

A gradient with decreasing content of acetonitrile was used and the non-polar components eluted in the void, while the polar were retained.

A slight baseline drift is hard to avoid at low UV detection, but they are not severe.

If you like we can run a sample, then contact us.
