I am looking for an LC/MS method to analyse Buprenorphin in human body fliuds. Can anyone help me with this problem?

We developed an LC-MS/MS method. It's quite difficult to achieve low LOQ as buprenorphine does NOT easily fragment in Q2. However, I am not allowed to give more details.

We recently developed and patented a new reverse stationary phase - Primesep D - the new column for direct injections of plasma and other biological samples. In brief, Primesep D is specifically designed for analysis of acidic, neutral and basic hydrophobic compounds (like Buprenorphine) in plasma containing analytes. In our testing, a sample prepared by diluting human plasma with water in 1:1 ratio was injected directly into the Primesep D column. The ACN-water-formic acid (0.2 %) has been used as a mobile phase. Our specially designed reverse stationary phase does not retain blood proteins, due to positive charge on a surface of the stationary phase and positive charge of serum albumin and most other bio-proteins at pH about 3.0. Proteins repel from the surface of the stationary phase and they are eluting in the void or before void. Small hydrophobic or negatively charged molecules retained in contrast. The retention of the acidic, hydrophobic neutral and basic compounds can be controlled by varying the amount of organic component (acetonitrile) in the mobile phase as on any RP column. A slight decrease in efficiency of the column is observed after 300 injections (10 ul each).
