I am developing a method for sucralose using RP-HPLC. Literature tells me to use C18 with Water/Organic. I have both BDS-C18 and Discovery RP-Amide C16.

When I test both columns under 70/30 Water/MeCN the Discovery C16 amide shows a lot more peak area for sucralose (factor of 50-100) at 190nm.

I have also tried with a Bonus-RP Amide and I get low area too.

Has anyone an explanation?

I have solved my problem - I had used TFA instead of hplc water in some of my work.
