Does it matter whether we use MeOH/Water or ACN/Water for needle wash? Which one is better? One of my co-worker said ACN will eat out rubber and thus ACN/Water will damage seals for injector. He said we should not use ACN/Water for needle wash even though our mobile phase has ACN in it. Is he right?

Dear AC

The seal materials in the injector of the 2690 are compatible with all common HPLC solvents (there are no rubber components), this includes both reverse and normal phase solvents. ACN/Water in the seal wash is an acceptable choice and will not damage the system in any way. The exact composition of the seal wash is dependent on your samples. You need enough organic in there to remove non-polar components and enough water to remove early eluters. The needle wash solvent (and plunger seal wash) solvents on the 2690 should be miscible with the mobile phase you are using.

I hope this answers all of your questions.

A.A.
