Methyl glucose

Hi all

I am wanting to analyse for methyl glucose with the existing equipment we have (Waters system with Alltech ELSD) We can analyse for the simple sugars with a MeCN:H2O mobile and NH2 C18 column, but would like to include methyl glucose as well.

Obviously i will try to elute Me-Glu with the existing mobile phase, but i would like to know if anybody has any expereince with Me-Glu analysis by LC?

My thoughts are to analyse the sample as is for the quantitation of glucose, then hydrolyse the sample, prior to re-analysis for glucose, the increase being equivalent to the orginal conc. of Me-Glu.

Can anyone give me any pointers here please??

I had analysed alfa-methyl glucoside, mono-, di and trimethyl methylglucosides some years ago. ODS column was found to be more adequate, each types of methylglucoside derivatives were separated well in low acetonitril containing aqueous mobile phase. The separation of anomers, however, is unsolved in this methods.

The separation of anomers can be manipulated with pH: at alkaline pH, they merge into a single peak. This should be doable with an XTerra reversed-phase column.

I have now been sucessful with the 250mm*4.6mm NH2 Alltima column and 1.5mL/min 80:20 MeCN:H2O with methyl glucose eluting at 5min and glucose at 7min, detection by ELSD.
