Fellow chromatographers:
I am trying to determine TRIS (tromethamine) in a pharmaceutical formulation. Can anyone give me some direction (column, mobile phase, detector, etc.)? I'm thinking RI detection because no chromophore but not sure. Thanks for any advice.

This is an ideal application for HILIC - i.e. a silica column with 70 to 80% acetonitrile and water, maybe with some buffer. It is not impossible that you see the compound in the low UV. I would try this first before using an RI, since it takes half a century for the baseline to settle in the RI, when you change the mobile phase.

Another alternative to HILIC can be mixed mode chromatography. TRIS can be analyzed same way as ethanolamines with ELSD detector. See example at our website

I'll try Uwe Neue's setup first as I don't have ready access to an ELSD detector. I'll keep you all posted.
