Can you calculate protein concentration from an A280 peak height without a pure protein standard, if you know the %solution extinction coefficient of the protein, i.e without a standard curve can you calculate protein concentrations from an HPLC.

What´s %solution extinction coefficient? If you have the extinction (better absorption) coefficient you can predict the peak hight via a complicated formular on page 87 of Snyder, Glajch, Kirkland, Wiley, 1988. From that approx. "calibration" you should be able to get at a concentration estimation. Or, more simply, use the coefficient to estimate the area as discussed in an earlier chain.
Personally, I use that only for a preliminary orientation.
