Dear Rajini,
Use SPE to extract the Vitamin D from the shark oil then use a 5 micron 250 x 4.6mm HyPurity Elite C18 (Hypersil)column or similar (Kromasil)under the following conditions :
Eluent : Methanol : Acetonitrile 30 : 70
Flow Rate : 1.0 ml/min
Detection : UV 254nm
Temperature : 40 degrees Celsius
Vitamin D2 elutes at 12.69 minutes
Vitamin D3 elutes at 13.40 minutes
All the best and goog luck with the chromatography.

Dear Thierry
Thank you very much. Do you know of any references for solid phase extraction? I haven't used it in the past.

Check out the Bakerbond or Varian Bond-elut SPE application notes or their bibliography. Or you can call their technical support for help.
