Help! Please suggest a HPLC/GC method for Vitamin D in Shark liver oil (anything other than that specified in the USP).

Dear Rajini,

Use SPE to extract the Vitamin D from the shark oil then use a 5 micron 250 x 4.6mm HyPurity Elite C18 (Hypersil)column or similar (Kromasil)under the following conditions :

Eluent : Methanol : Acetonitrile 30 : 70
Flow Rate : 1.0 ml/min
Detection : UV 254nm
Temperature : 40 degrees Celsius

Vitamin D2 elutes at 12.69 minutes
Vitamin D3 elutes at 13.40 minutes

Thank you very much. Do you know of any references for solid phase extraction? I haven't used it in the past.
