Although all my HPLC experience is with proteins and peptides, I have been asked to see about developing an assay to compare batches of sybr green stain, with the intent of figuring out why some lots of the stain work in our application and why some don't. Can I get suggestions on appropriate column types, HPLC detector, solvents, sample prep, etc? Or maybe some suggested resources such as good web sites, books, or column selection guides? Thank you -

A C18 column, a UV/Vis detector, dissolve the sample in water and run a gradient as you would do for a peptide (0.1% TFA, from 95% water to 95% acetonitrile). See where the green stuff elutes, and run a flatter gradient around this solvent composition. The same thing as you would do for a peptide separation...
