hello everybody
we are doing amino acid analysis by HPLC after OPA/Fmoc derivatization (similar to Aminoquant)
do you know of a nice method to end up the system during week-ends or after a day sequence ?

Do you mean to stop the machine?

I used to create a programe of finish with zero mL/min flow. Then, you programe another secuence with no sample.

If it's not what you ask, please tell me.

not to stop the machine or the pump, the Agilent software has a function to do this
I am looking for a method to rinse the column and store it

You should wash out the buffer salts by flushing
the column with a mixture of your organic
modifier and pure water (e.g. 50:50) with at
least 10 column volumes. Then you can run the
system with very low flow (0,02 ml/min) during
weekend.
