I am trying to establish a HPLC method for determining Ascorbic acid and its derviatives (i.e. ascorbic acid-2-phosphate) in tissue culture medium. Does anyone have any experience in this area?

For trace amounts of ascorbic acid in a "soup" you should look into electrochemical(amperometric) detection.
