In case of related substances detemination, is it necessary to take the moisture content of working standard before use for hygroscopic material and then use the corrected potency for calculations?

When using a reference standard, the standard should be well characterized and the purity known. For example, hypothetically, if your standard is "100% pure" in the sense that there are no other compounds except it contains 1.5% water, the purity of your standard is 100%-1.5% = 98.5%. Your calculation must take this into account.
