I'm using a Zorbax C8 cartridge column with a guard column attached. Mobile phase is 100% ACN.

What is the best sample to evaluate t0, the column dead time?

I was using 1 mM uracil, but the "unretained" peak seems to tail, and there is also a concentration dependence on the Rt, implying nonlinear chromatography. Would NaNO3 be a better sample to measure t0?

Spike your sample with a touch af acetone. It is unretained and is easily detected by UV (unlike NaNO3). Based on your column and mobil phase, you could pick from a large variety of compounds because little is going to be retained.
