I'm trying to remove strongly retained components from a Zorbax Rx C8 column. Standard MP is 100% ACN. I'm currently washing the column for 30 column volumes with 0.1% TFA in ACN. This works okay, but I'm still having reproducibility problems.
Can I safely increase the TFA concentration to 0.2% or higher? What are some other methods for column cleaning...without resorting to extreme measures (backflushing, etc)?

Depends what's on there. Essentially, you want to use something which is a good solvent for the contaminants. I've seen lipids/waxes taken off with methylene chloride, proteins taken off with guanidine hydrochloride or isopropanol, etc. Just make sure that whatever you use is miscible with what you were using and allow sufficient column washout (10-20 column volumes).
