Reducing Column Diameter in Gradient Elution -- A Case Study

Simply scaling the gradient won't always work

Keywords: column diameter, narrow-bore columns, dwell volume, mixer volume

Erratum:

Figure 3 and the accompanying discussion is not quite correct. The correct numbers are (assuming that a 150 x 4.6 mm column has a column volume, Vm = 1.5 mL and a 150 x 2.1 mm column has Vm = 0.3 mL):

3a: 1  min dwell time = 0.66 Vm dwell; 15 mL gradient = 10 Vm gradient
3b: 2.5 min dwell = 3.3 Vm; 4 mL gradient = 13 Vm
3c: 5 min dwell = 3.3 Vm; 2 mL gradient = 6.6 Vm
